چکیده

Salvia hypoleuca is used in Iranian traditional medicine as an agent for treatment of some diseases and troubles. In this study, attention was focused on the antioxidant effect of Salvia hypoleuca. The aim of the present study was to determine the effects of Salvia hypoleuca extract on Endotoxin-Induced Uveitis (EIU) in rats. In addition, the endotoxin-induced expression of the inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 proteins was determined in a mouse macrophage cell line (RAW 264.7 ) treated with Salvia hypoleuca extract in vitro, to clarify the anti-inflammatory properties. EIU was induced in male Wistar rats by a footpad injection of lipopolysaccharide (LPS). Immediately after the LPS injection, 1, 10, or 100 mg extract or 10 mg prednisolone was injected intravenously. After 24 h, the aqueous humor was collected from both eyes, and the number of infiltrating cells, protein concentration, Nitric Oxide (NO), prostaglandin (PG)-E2, and TNF-alevels in the aqueous humor were determined. RAW 264.7 cells treated with various concentrations of extract were incubated with 10 :g/mL LPS for 24 h. Levels of NO, PGE2, and TNF-a were determined. The expression of iNOS and COX-2 proteins was analyzed by Western blot analysis. The number of inflammatory cells, the protein concentrations, and the levels of NO, PGE2, and TNF-ain the aqueous humor in the groups treated with Salvia hypoleuca were significantly decreased in a dose-dependent manner. In addition, the anti inflammatory effect of 100 mg Salvia hypoleuca was as strong as that of 10 mg prednisolone. The antiinflammatory action of Salvia hypoleuca was stronger than that of either quercetin or anthocyanin administered alone. Extract also suppressed LPS-induced iNOS and COX-2 protein expressions in RAW 264.7 cells in vitro in a dosedependent manner. The results suggest that Salvia hypoleuca has a dose-dependent anti-ocular inflammatory effect that is due to the direct blocking of the expression of the iNOS and COX-2 enzymes and leads to the suppression of the production of NO, PGE2, and TNF-a.

Intra-vitreal injection of methotrexate in experimental endotoxin-induced uveitis in rabbit

Uveitis is a major cause of vision loss. Methotrexate (MTX) has been widely used in uveitis due to its relatively safe profile. The purpose of this study was to evaluate the effects of two different dosages of MTX via intra-vitreal administration for treatment of endotoxin induced uveitis (EIU) in an experimental model. Thirty-five healthy rabbits were randomly divided into four groups and all ...

Anti-inflammatory effects of aronia extract on rat endotoxin-induced uveitis.

PURPOSE Aronia crude extract (ACE) with high levels of polyphenol compounds has been reported to have antioxidative effects in vitro and in vivo. In this study, attention was focused on the antioxidant effect of ACE. The purpose of the present study was to investigate the effect of ACE on endotoxin-induced uveitis (EIU) in rats. In addition, the endotoxin-induced expression of the inducible nit...

Cytokines: the good, the bad, and the unknown.

tiight decades ago, a leading authority on uveitis hypothesized that many patients with uveitis suffered from a disease induced by bacterial toxins. Paradoxically, bacterial toxin in the form of endotoxin evolved as a conventional therapy for uveitis."" Now the conundrum has come full circle; currently, a subcutaneous or intraperitoneal injection of endotoxin serves as a standard model to induc...

Green Tea Extract Treatment Alleviates Ocular Inflammation in a Rat Model of Endotoxin-Induced Uveitis

Green tea extract (GTE) ingested by rats exerted anti-oxidative activities in various ocular tissues as shown in our previous studies. The present work investigated anti-inflammatory effects of GTE on endotoxin-induced uveitis (EIU). EIU was generated in adult rats by a footpad injection of 1 mg/kg lipopolysaccharide (LPS). Oral administration of GTE (550 mg/kg) was given one, two or four times...

Relative potency of tyrosol in the treatment of endotoxin-induced uveitis in rats

Tyrosol (Tyr) is a natural phenolic antioxidant with diverse biological activities. We compared the anti-inflammatory effects of intravenously administered Tyr versus prednisolone (PSL) in an endotoxin-induced uveitis (EIU) rat model. Intravenous administration of 100 mg/kg Tyr was performed 2 hr before, simultaneously and 2 hr after lipopolysaccharide (LPS) injection. Tyr treatment was associa...
